Could your evening glass of wine trigger Alzheimer’s? What does a shocking new study say about alcohol use disorder?

A recent study from Scripps Research has revealed an intriguing connection between Alzheimer’s disease and alcohol use disorder, shedding light on how excessive drinking can accelerate the progression of this devastating neurodegenerative condition. Published in eNeuro, the findings highlight shared disruptions in inflammation and cell-death pathways in the brain and provide deeper insight into the molecular relationships between these two conditions.

Increasing impact of Alzheimer’s disease

Alzheimer’s disease is the leading cause of dementia, affecting approximately 7 million Americans – a number expected to double by 2060. While aging and genetics are important contributors, lifestyle factors such as alcohol consumption are increasingly being recognized as playing a role in the onset and progression of the disease.

Heavy drinking has long been linked to cognitive decline. But, this new study delves deeper into the biological mechanisms, suggesting that similar molecular disruptions may occur in alcohol use disorders and Alzheimer’s disease.

conclusion

Researchers led by Dr. Arpita Joshi analyzed gene expression patterns in the brains of individuals at different stages of Alzheimer’s disease. Using advanced techniques such as single-cell transcriptomics, they focused on the neocortex – a brain region important for memory and reasoning.

The study included brain samples from 75 individuals with Alzheimer’s and compared them to a smaller dataset of individuals with alcohol use disorder. The goal was to identify overlapping genetic pathways that could explain the similarities between the two conditions, and they discovered –

  • Inflammation and immune response: In both Alzheimer’s disease and alcohol use disorder, activation of inflammatory genes was observed, particularly in microglia (immune cells of the brain) and vascular cells. This inflammation can disrupt the blood-brain barrier, allowing harmful substances to affect brain function.
  • Neurological Impairment: Both conditions were associated with decreased gene activity related to synaptic signaling necessary for communication between brain cells. This impairment was particularly pronounced in individuals with advanced Alzheimer’s stages and alcohol dependence.
  • vascular disease: Disruption of genes related to blood vessel growth and maintenance was observed, suggesting shared damage to the brain’s vascular system.
  • Stage-specific changes: In the early stages of Alzheimer’s, the brain shows changes in autophagy, a process that helps clear damaged cellular components. In later stages, inflammation, cell death, and disruptions in brain signaling become more prominent.

role of alcohol

The overlap between the two conditions points to alcohol use disorder as a potential risk factor for progression to Alzheimer’s. Alcohol appeared to exacerbate some of the same molecular disruptions seen in Alzheimer’s, particularly in genes related to inflammation and neuron function.

“Alcohol use disorder and Alzheimer’s disease have significant similarities in terms of the disorders they cause, suggesting a potentially serious impact of alcohol use disorder on the progression of Alzheimer’s disease,” Dr. Joshi said.

Why does it matter?

Understanding these shared pathways provides new avenues for prevention and treatment. If alcohol use accelerates the progression of Alzheimer’s, reducing consumption may be a modifiable way to reduce risk.

It also underlines the importance of brain health when making lifestyle choices. Although occasional drinking may not pose any significant risk, excessive or prolonged alcohol consumption can have lasting effects on cognitive health.

boundaries

The study had some limitations, particularly the small dataset for alcohol use disorder. Further research with larger, more diverse populations is needed to confirm these findings and explore how molecular changes translate into brain function and behavior.

Dr. Joshi and his team are already working on a more comprehensive alcohol use disorder dataset to better understand how drinking affects the brain and its potential connection to Alzheimer’s.

This study serves as a powerful reminder that our lifestyle choices play an important role in shaping our long-term health. By understanding the risks and taking proactive steps, we can protect our brain and maintain cognitive vitality as we age.
